Available studies in humans and postmortem analysis of tissues suggested that the oral bioavailability of titanium dioxide in humans is very low. JECFA noted that there are currently no epidemiological studies that allow any conclusions to be drawn with respect to an association between dietary exposure titanium dioxide and human health effects.

Faber argued there hasn't been enough change in these federal regulations in the decades following the FDA's approval of titanium dioxide – especially as others increasingly point to potential health consequences.

There is some evidence that ingested titanium dioxide does not completely exit the body. A 2015 review of animal studies and a few human studies suggests titanium dioxide can get absorbed into the bloodstream and expose other organs to damage.

The decrease in the immune response could be the result of damage to the spleen, which is the largest immune organ in animals and plays an important role in the immune response. Powel et al. demonstrated that TiO₂ NPs may trigger immune reactions of the intestine after oral intake. They showed that TiO₂ NPs conjugated with bacterial lipopolysaccharide, but not TiO₂ NPs or lipopolysaccharide alone, trigger the immune response in human peripheral blood mononuclear cells and in isolated intestinal tissue. This indicates that TiO₂ NPs may be important mediators in overcoming normal gut-cell hyporesponsiveness to endogenous luminal molecules, which may be particularly relevant to patients with inflammatory bowel disease, which is characterized by an abnormal intestinal permeability.

In industrial settings, people can be exposed to titanium dioxide through inhalation. Inhalation exposure to titanium dioxide is exceedingly rare for most people. 

The EU expert panel did not identify an immediate health concern linked to TiO₂ when used as a food additive. However, due mainly to uncertainties concerning the safety of TiO₂ nanoparticles, the panel concluded that TiO₂ as a food additive (E171) could no longer be considered safe.

Mars Wrigley, the company that makes Skittles, is being sued by a California man who claims the candy contains a known toxin that poses such a serious health risk that Skittles are unfit for human consumption.

Although the evidence for general toxic effects was not conclusive, on the basis of the new data and strengthened methods we could not rule out a concern for genotoxicity and consequently we could not establish a safe level for daily intake of the food additive, commented Matthew Wright, member of the EFSA's Food Additives and Flavourings Panel in a press statement.

Resumen–En este artículo se discute el descubrimiento del litopón fosforescente en dibujos a la acuarela por el artista americano John La Farge, fechados de 1890 a 1905, y la historia del litopón en la industria de los pigmentos a finales del Siglo XIX y principios del Siglo XX. A pesar de tener muchas cualidades deseables para su uso en pintura para acuarela o pinturas al óleo blancas, el desarrollo del litopón como pigmento para artistas fue obstaculizado por su tendencia a oscurecerse con la luz solar. Su disponibilidad para los artistas y su adopción por ellos sigue siendo poco clara, ya que por lo general los catálogos comerciales de los coloristas no eran explícitos al describir si los pigmentos blancos contenían litopón. Además, el litopón se puede confundir con blanco de plomo durante el examen visual, y su fosforescencia de corta duración puede ser fácilmente pasada por alto por el observador desinformado.
